Application of a Microfluidic Droplet Mixer for [18f] Fluorine Labeling of Biomolecules for Positron Emission Tomography

چکیده
The most widely used method to label biomolecules with fluorine-18 is the formation of an amide bond between a lysine residue and 4-fluorobenzoate by reaction with N-succinimidyl 4-[F]fluorobenzoate ([F]SFB). The reaction of [F]SFB with the biomolecule is usually performed in macroscopic scale, whereas the reagents are only present in microscopic amounts. Thus, it could be beneficial to carry out such reactions in a microfluidic environment. We developed a microfluidic droplet mixer, which is capable of mixing two solutions quickly. A chemical reaction can be performed thereafter while the mixture flows through a long channel.

Positron emission tomography (PET) using radiolabeled biomolecules is a translational molecular imaging technology that is increasingly used in support of drug development. Current methods for radiolabeling biomolecules with fluorine-18 are laborious and require multistep procedures with moderate labeling yields. The Al18F-labeling strategy involves chelation in aqueous medium of aluminum mono[...

Positron Emission Tomography (PET) has become a popular imaging technique widely used for diagnostic purposes. To date, much attention has been devoted to 18F-fluoride because of the characteristics of its nuclear decay, as well as its relative ease of preparation from 18O-water. However, with a half-life of 110 minutes, swift and efficient incorporation of 18F-fluorine into biomolecules is nec...
